Fluorouracil

Term · Oncology and biomedicine · MLC-T-ONC-003273

  1. A drug given as an injection to treat cancers of the breast, colon, rectum, stomach, and pancreas and as a cream to treat actinic keratosis (a skin condition that may become cancer) and certain types of basal cell skin cancer. It is used under the brand names Carac, Tolak, Efudex, and Fluoroplex to treat actinic keratosis. It is also used under the brand name Efudex to treat basal cell skin cancer. Fluorouracil is also being studied in the treatment of other conditions and types of cancer. It stops cells from making DNA and may kill cancer cells. Fluorouracil is a type of antimetabolite. Also called 5-fluorouracil and 5-FU.

  2. An antimetabolite fluoropyrimidine analog of the nucleoside pyrimidine with antineoplastic activity. Fluorouracil and its metabolites possess a number of different mechanisms of action. In vivo, fluoruracil is converted to the active metabolite 5-fluoroxyuridine monophosphate (F-UMP); replacing uracil, F-UMP incorporates into RNA and inhibits RNA processing, thereby inhibiting cell growth. Another active metabolite, 5-5-fluoro-2'-deoxyuridine-5'-O-monophosphate (F-dUMP), inhibits thymidylate synthase, resulting in the depletion of thymidine triphosphate (TTP), one of the four nucleotide triphosphates used in the in vivo synthesis of DNA. Other fluorouracil metabolites incorporate into both RNA and DNA; incorporation into RNA results in major effects on both RNA processing and functions.
