Histrelin acetate

  1. A drug used under the brand name Vantas as palliative therapy for advanced prostate cancer. Histrelin acetate initially causes the pituitary gland to make more luteinizing hormone (LH) and follicle-stimulating hormone (FSH), temporarily increasing testosterone levels in men. With continued use, histrelin acetate lowers the amount of LH and FSH the pituitary gland releases, leading to a drop in testosterone levels. Histrelin acetate is also used under the brand name Supprelin LA to treat early puberty in males and females. It is a type of hormone therapy called a luteinizing hormone-releasing hormone (LHRH) agonist and a type of androgen deprivation therapy that is reversible.

  2. The acetate salt form of histrelin, a long-acting, synthetic nonapeptide analog of gonadotropin-releasing hormone (GnRH) with potential anti-tumor activity. Upon administration, histrelin binds to and activates GnRH receptors; prolonged administration results in pituitary GnRH receptor desensitization and inhibition of follicle stimulating hormone (FSH) and luteinizing hormone (LH) secretion, leading to a significant decline in testosterone production in males and may inhibit androgen receptor-positive tumor progression; in females, prolonged administration results in decreased estradiol production.
