Revumenib citrate

Term · Oncology and biomedicine · MLC-T-ONC-007420

  1. A drug used to treat adults and children aged 1 year and older with acute leukemia that has a certain change in the KMT2A gene and has come back or has not gotten better with other treatment. It is also being studied in the treatment of other types of cancer. Revumenib citrate binds to a protein called menin and keeps it from binding to another protein called KMT2A. This stops or slows the growth of leukemia cells with changes in the KMT2A gene. Also called Revuforj.

  2. The monocitrate monohydrate form of revumenib, an orally bioavailable protein-protein interaction (PPI) inhibitor of menin with both wild-type and fusion proteins of lysine methyltransferase 2A (mixed lineage leukemia MLL; myeloid/lymphoid leukemia; KMT2A), with potential antineoplastic activity. Upon oral administration, revumenib targets and binds to the nuclear protein menin, thereby preventing the interaction between the two proteins menin and KMT2A and the formation of the menin-KMT2A complex. This reduces the expression of downstream target genes and results in an inhibition of the proliferation of KMT2A-rearranged leukemic cells. The menin-KMT2A complex induces the transcription of various genes involved in the survival, growth, transformation and proliferation of certain kinds of leukemia cells.
