Peptide de novo sequencing

The process of determining the amino acid sequence of a peptide directly from its tandem mass spectrometry (MS/MS) data without reference to a protein or DNA database. The peptide is fragmented within the mass spectrometer, and the resulting spectrum of fragment ions is interpreted to reconstruct the sequence. The primary data used are the mass differences between adjacent peaks in the b-ion or y-ion series, which correspond to the residue masses of individual amino acids.
