Stable isotope ratio analysis of amino acids in cell culture

A metabolic labelling approach in which cultured cells grow in medium containing amino acids enriched in stable isotopes, such as 13C- or 15N-labelled lysine and arginine, so that newly made proteins carry a known mass shift. Labelled and unlabelled cell populations are mixed and analysed by mass spectrometry, and peptide peak intensity ratios give relative protein abundance. Heavy and light forms of a labelled peptide typically differ by several daltons.
