Isocratic analysis
Term · Chemistry · MLC-T-CHM-006174
A chromatographic separation carried out with a mobile phase whose composition is held fixed from injection to the end of the run, in contrast to gradient elution, where the proportions of the solvents are changed during the run. Because no gradient program or re-equilibration is needed, the method is simple to set up, and it suits samples whose components differ little in polarity. Reversed-phase separation of a few related compounds on a C18 column with a fixed 60:40 methanol and water eluent is a typical case.
