A three-dimensional nuclear magnetic resonance spectroscopy experiment that correlates the chemical shift of an amide proton and its attached nitrogen with the alpha carbon of the preceding amino acid residue. Magnetization is transferred from the amide proton to its nitrogen, then across the peptide bond to the preceding carbonyl carbon, and finally to the preceding alpha carbon. This experiment provides sequential assignment information by linking adjacent residues in a polypeptide chain.
