Nuclease assay

A method for detecting or measuring a specific target nucleotide sequence by using a nuclease to cut nucleic acid and reading the result. In the 5' nuclease assay of real-time PCR, the polymerase's exonuclease activity cleaves a probe bound to the target and frees a fluorophore, while in nuclease protection assays, a probe annealed to the target survives digestion of unbound nucleic acid.
