Observation volume

The specific three-dimensional region within a sample or analytical device from which a detectable signal originates during a measurement. This volume is defined by the intersection of the excitation source and the detection optics, determining the amount of material contributing to the signal. In fluorescence spectroscopy, for example, it is the volume where excitation light is absorbed and emission is collected.
