Radial immunodiffusion

A quantitative immunoassay in which antigen placed in a well diffuses into agar containing a specific antibody and forms a ring of precipitate where the two reach equivalence. In the endpoint (Mancini) method the square of the ring diameter is proportional to antigen concentration, so readings from standards give a calibration line for unknowns. Serum immunoglobulins and complement components were routinely measured this way before nephelometry largely replaced it.
