Charge site derivatization

A chemical modification of an analyte to introduce a permanent, localized charge at a specific location within the molecule. This derivatization directs fragmentation in tandem mass spectrometry by creating a fixed charge site, which is not the mobile proton that typically initiates fragmentation pathways. For example, the quaternization of a tertiary amine with an alkyl iodide introduces a fixed positive charge on the nitrogen atom, altering subsequent fragmentation patterns.
