Immobilised metal ion affinity-based fluorescence polarisation assay

A fluorescence polarization assay for kinase and phosphatase activity in which a fluorescently tagged peptide substrate, once phosphorylated, binds through its phosphate group to nanoparticles coated with trivalent metal ions. Binding to the large nanoparticle slows the tumbling of the peptide, so its fluorescence polarization rises, while the unphosphorylated peptide stays free and shows low polarization. Kinase activity therefore raises the signal, whereas phosphatase activity lowers it.
