Orthosteric site

The primary binding site on a receptor where the endogenous ligand normally interacts to produce a physiological response. Drugs that bind to this site are called orthosteric ligands and can act as agonists or antagonists, directly competing with the natural ligand. This site is distinct from allosteric sites, which are located elsewhere on the receptor.
