Total internal reflection fluorescence

A microscopy technique that selectively illuminates and excites fluorophores within a very thin region (typically 100-200 nanometers) immediately adjacent to the interface between two media with different refractive indices. An evanescent wave, generated by total internal reflection of an incident laser beam, penetrates only a short distance into the sample, minimizing background fluorescence from the bulk solution. This method is particularly effective for studying surface phenomena and single-molecule dynamics.
