Two-site immunoradiometric assay

A type of immunoradiometric assay in which two antibodies bind different epitopes of the same antigen molecule and only one of them is labelled with a radionuclide. The unlabelled antibody is usually immobilized on a solid phase and captures the antigen, and the labelled antibody then binds the captured antigen, so the bound radioactivity rises with antigen concentration. The sandwich format gives high specificity because both antibodies must recognise the antigen.
